ABSTRACT:
Influenza virus remains a major global health threat, causing millions of infections and hundreds of thousands of deaths annually. The emergence of resistance to conventional antiviral drugs, including oseltamivir, underscores the need for novel therapeutic options. This study evaluates the potential of three olive-derived phytoconstituents—oleuropein, verbascoside, and hydroxytyrosol—as inhibitors of influenza virus neuraminidase. Advanced molecular docking revealed that verbascoside exhibited the highest binding affinity (-8.9 kcal/mol), surpassing oseltamivir (-5.5 kcal/mol), indicating superior neuraminidase inhibition. Pharmacokinetic assessments using SwissADME and pkCSM demonstrated favorable drug-likeness, absorption, and metabolic profiles for these compounds, supporting their therapeutic viability. Olive polyphenols are known for their anti-inflammatory, antioxidant, and cardioprotective properties, this study highlights their potential as natural anti-influenza agents with enhanced binding efficacy compared to standard antivirals. These findings suggest that olive-derived compounds, particularly verbascoside, may serve as promising alternatives for combating influenza, especially in cases of antiviral resistance.
